摘要
建立了同时测定大叶白麻中芦丁、金丝桃苷、槲皮素含量的HPLC分析方法。色谱条件为,色谱柱:KromasilC18(5μm,4.6mm×250mm),流动相∶甲醇-0.04%磷酸水线性梯度洗脱,柱温:30℃,流速:1.0mL/min,检测波长:360nm。结果表明,芦丁、金丝桃苷和槲皮素分别在2.7~13.3μg、0.3~2.3μg、2.4~14.4μg范围内,进样量与色谱峰积分面积呈良好的线性关系,相关系数分别为0.9996、0.9999、0.9999,平均回收率(n=6)分别为100.45%、100.26%、98.92%。本法可同时测定芦丁、金丝桃苷、槲皮素的含量,且稳定可靠,重复性好,可作为大叶白麻有效成分分析的有效手段。
In this paper, the content of rutine, hyperin and quercetin in Poacynum hendersonii (Hook.f.) Woodson were detected by HPLC which separated on the Kromasil C18 (5μm,4.6mm×250mm) column with 0.04% phosphoric acid (gradient elution) at 30℃. The flow rate was 1.0 mL/min and detected at 360nm. The results revealed that rutine was proved to be linear in the range of 2.7~13.3 μg with a regression coefficient of 0.999 6, its average recovery was 100.45%. Hyperoside was proved to be linear in the range of 0.3 ~2.3 μg with a regression coefficient of 0.999 9, its average recovery was 100.26%. Quercetine was proved to be linear in the range of 2.4 ~14.4 μg with a regression coefficient of 0.999 9, its average recovery was 98.92%. As a results, this method was able to detect rutine,hyperin and quercetin simultaneously, quickly and reliably.
