β淀粉样蛋白(1-40)对痴呆老龄大鼠海马MARCKS mRNA表达及学习记忆的影响 被引量:3

摘要 目的:研究β-淀粉样蛋白(1-40)致痴呆老龄大鼠海马富含丙氨酸的豆蔻酰化蛋白激酶C的作用底物(MARCKS mRNA)的表达和学习记忆功能的影响。方法:将大鼠随机分为年轻组、老龄组、假手术组以及模型组,对老龄大鼠采用β-淀粉样蛋白(1-40)1次性侧脑室注射,建立痴呆模型。通过行为学以及RT-PCR检测,观察模型大鼠学习记忆功能以及海马MARCKS mRNA的变化。结果:行为学检测表明模型组的学习记忆功能明显低于正常组,模型组MARCKS mRNA表达明显高于老龄组、正常组及假手术组均明显降低(P<0.01)。结论:侧脑室注射β-淀粉样蛋白(1-40)可以明显增加老龄大鼠海马MARCKS mRNA的表达,并且导致老龄大鼠学习记忆障碍。提示MARCKS表达异常很可能是β-淀粉样蛋白(1-40)致神经元可塑性降低的关键环节。
