野生型p53基因转染U14细胞增强肿瘤抗原的免疫原性 被引量:1

摘要 目的:探讨p53基因修饰的U14细胞瘤苗能否增强抗小鼠宫颈癌U14细胞的主动免疫反应。方法:随机将实验分3组:p53-U14细胞瘤苗组、U14细胞瘤苗组、空白对照组;运用阳离子脂质体介导的转染方法,将p53基因导入U14细胞,用含G418的选择性培养基对已转染细胞进行筛选,以获得稳定表达相应P53蛋白的U14细胞,用间接免疫细胞化学方法检测P53蛋白的表达情况。采用反复冻融法分别制备p53-U14细胞瘤苗和U14细胞瘤苗,在此基础上采用MTT法检测经各组细胞瘤苗免疫后的小鼠脾细胞对U14细胞的细胞毒作用。结果:效靶比为20:1时,p53-U14细胞瘤苗组、U14细胞瘤苗组和空白对照组刺激的脾细胞杀伤活性分别为64.91%、42.14%和21.66%;效靶比为40:1时,p53-U14细胞瘤苗组、U14细胞瘤苗组和空白对照组刺激的脾细胞杀伤活性分别为58.87%、42.91%、8.19%。结论:p53基因修饰的U14细胞确能提高肿瘤抗原的免疫原性。
