全反式维甲酸对不同增殖潜能的结肠癌细胞株HIF-1α表达的影响 被引量:5

摘要 目的:探讨全反式维甲酸(ATRA)对不同增殖结肠癌细胞系的生长抑制作用,以及对低氧诱导因子(HIF-1α)表达的抑制作用。方法:采用体外培养属于结肠癌DukesC期的HCT-8细胞和LOVO细胞,不同浓度的全反式维甲酸(ATRA)干预。利用MTT观察ATRA对结肠癌细胞系的生长,采用半定量RT-PCR和Western印迹检测ATRA干预后在低氧状态下结肠癌细胞中HIF-1α的表达。结果:全反式维甲酸(ATRA)对结肠癌细胞的增殖有抑制作用,对不同增殖能力的结肠癌细胞HIF-1α的表达量有抑制作用。结论:全反式维甲酸可以抑制结肠癌细胞的增殖,对结肠癌细胞HIF-1α的表达有明显的抑制作用。
