18三体及假肥大型肌营养不良成纤维细胞系的建立

摘要 目的:建立18三体及假肥大型肌营养不良胎儿皮肤成纤维细胞系。方法:3名在我院进行羊水染色体诊断或基因诊断,诊断为异常的胎儿。获得知情同意后,取1小块皮肤,进行成纤维细胞培养及鉴定。结果:对皮肤进行分离、培养,成功建立了3株疾病皮肤成纤维细胞系,包括1株18三体、2株假肥大型肌营养不良,并进行了传代、冻存、复苏及鉴定。在传代培养过程中核型稳定,STR基因检测对这些细胞系进行个体识别。结论:本研究在体外建立了稳定的18三体及假肥大型肌营养不良胎儿皮肤成纤维细胞系,为进一步研究相关疾病提供充足的细胞来源。
