摘要
目的检测人胶质瘤与非肿瘤脑组织中微小RNA(miRNA)的表达差异情况。方法选取经手术切除的胶质瘤组织标本及非肿瘤脑组织,分别提取总RNA和小分子RNA,在miRNA微阵列芯片中杂交检测,通过芯片扫描和数据聚类分析,并通过实时荧光定量RT-PCR验证,获得该病异常miRNAs表达谱。结果筛选出5个胶质瘤的相关miRNAs,其中表达上调2个,表达下调3个。其中上调的有miR-18b、miR-486;下调的有miR-237、miR-96、miR-21,差异表达情况得到定量RT-PCR的证实。结论筛选出的差异表达miRNAs可能参与胶质瘤的发生发展。
Objective To explore the differentially expressed microRNAs in glioblastoma and to the relationship between pathogenesis using miRNA microarray.Methods The tumor tissue glioblastoma and non-tumors tissue were obtained from undergoing surgical resection.Total RNA,including the small RNAs,was extracted and isolated respectively.Hybridization was carried out on miRNA microarray chip.Real-time quantitative RT-PCR was performed to confirm results obtained by microarray analysis.Results Five miRNAs have been identified,2 of them were up-regulated and the remaining three were down-regulated in cancer tissues.The differential expression of miR-18b、miR-486、miR-237、miR-96、miR-21 was confirmed by microarray analysis.Conclusion The differentially expressed microRNAs miRNA genes in this study may be involved in the pathogenesis of human glioblastoma.
