摘要 目的探讨雷公藤甲素(TP)对T细胞转录因子核因子-κB(NF-κB)和活化蛋白-1(AP-1)的DNA结合活性的作用。方法通过凝胶电泳迁移试验(EMSA)检测TP对伴刀豆球蛋白A(ConA)或佛波脂(PMA)刺激活化的人T细胞NF-κB、AP-1的DNA结合活性的影响。结果正常T细胞NF-κB、AP-1的DNA结合活性较弱,经ConA或PMA刺激后结合活性增强,TP可降低NF-κB、AP-1的DNA结合活性。结论 TP抑制NF-κB、AP-1的DNA结合活性,可能是其免疫抑制作用的重要机制。 Objective To investigate the effects of triptolide(TP) on DNA binding activities of nuclear factor-κB(NF-κB) and activator protein-1 (AP-1) of T cells. Methods The effects of TP on DNA binding activities of NF-κB and AP-1 of human T cells were stimulated by concanava]in A(ConA) or phorbol myristate acetate(PMA) , then nuclear protein were extracted and the DNA binding activities of NF-κB and AP-1 were detected by eletrophoretic mobility shift assay(EMSA). Results The DNA binding activities of NF-κB and AP-1 of normal T cells were weaker, which were stronger after stimulated by ConA or PMA. TP could inhibit DNA binding activities of NF-κB and AP-1. Conclusion One possible important mechanism of immunosuppressive effect of TP is that TP inhibits DNA binding activities of NF-κB and AP-1.
