摘要
分离提纯是制备功能性木低聚糖的关键技术之一。本文利用聚丙烯酰胺凝胶(Bio-GelP-4)柱层析技术能将木低聚糖混合样品较好地分离开来。当分离条件为:层析柱尺寸Φ3.0×120cm,去离子水为洗脱液,流速25ml/h,柱温50℃,可以得到木二糖至木四糖的单一组分。研究表明,采用AminexHPX-42A糖柱是测定低聚糖的简便快速的好方法。此外,在无高效液相设备的条件下,通过DNS法测定还原糖浓度,也可以初步判断低聚糖的分离效果,不失为一种简单、快速和经济的分析方法。
Xylooligosaccharides with specific biological activities have been paid great attention. This study found that xylooligosaccharides could be isolated effectively on a polyacrylamide (BioGel P4) column and some pure samples such as xylobiose, xylotriose and xylotetraose were obtained. The suitable separating conditions were: column size, 3.0120cm; mobile phase, deionized water; flow rate, 25ml/h; column temperature, 50. In addition, xylooligosaccharides could be determined by HPLC on Aminex HPX42A column excellently.
