摘要
采用PCR-SSCP法对白羽番鸭脂联素基因内含子进行SNP检测,并分析SNP对肉质的遗传效应。结果表明,检测到3种基因型(CC、CT和TT)和2个等位基因(C和T),CT和C分别为优势基因型和优势等位基因。序列比对发现,在内含子内存在1个SNP位点T290C,该位点在白羽番鸭群体中表现为中度多态,处于Hardy-Weinberg平衡状态(P>0.05);最小二乘法分析表明,CC基因型个体的IMF显著高于TT基因型个体(P<0.05),失水率极显著低于TT基因型个体(P<0.01);CT基因型个体的失水率显著低于TT基因型个体(P<0.05)。结果揭示,T290C位点可能对脂肪沉积有显著影响,CC基因型是白羽番鸭肉质较为优良的基因型。
The full-length of adiponectin gene intron of White Muscovy was amplified using speci-fic primers designed from Cairina moschata DNA sequence,and the polymorphism were detected by PCR-SSCP method and its association with meat quality were further analyzed.Two alleles(C and T) and 3 genotypes(CC,CT and TT) were observed,among which C and CT were predominant.One SNP(T290C) was found in the intron of adiponectin gene.The T290C loci belongs to moderate polymorphism and is in accordance with Hardy-Weinberg equilibrium(P〉0.05).IMF of the birds of genotype CC are significant higher than that of genotype TT(P〈0.05).Percentage of chest muscle loss water of the birds with genotype CC and genotype CT are extremely significant or significant lower than that of genotype TT(P〈0.01 or P〈0.05).These results suggest that the T290C loci has genetic effect for fatty deposition and genotype CC is the helpful genotype of meat quality in White Muscovy.
