摘要 目的建立反相高效液相色谱法测定杯苋甾酮在家兔血浆中的药物浓度。方法血浆样品用乙腈提取,流动相为乙腈20g、水80g、磷酸二氢钠1.20g,流速1mL/min,检测波长243nm。结果家兔口服给药后血浆中可检出杯苋甾酮,杯苋甾酮在0.565~9.040μg/mL范围内线性关系良好,r=0.9990(n=5),日内RSD为±3.9%,日间RSD为±7.3%;血浆最低检测浓度为0.28μg/mL。结论本法快速、准确、重现性好,为该药物的药代动力学研究提供依据。 Objective To establish a RP-HPLC(reversed-phase high-performance liquid chromatography) method for the determination of cyasterone in rabbit plasma.Methods The plasma samples were extracted with Methyl Cyanides.RP-HPLC conditions were set as follows:Mobile phase:CH3CN 20 g、H2O 80 g、NaH2PO4 1.20 g;Flow rate:1 mL/min,Detection wave length:243 nm.Results Cyasterone was found in rabbit plasma after oral drug administration.The calibration curve of cyasterone showed good linearity over the range of 0.565~9.040 μg/mL,r=0.999 0(n=5).RSD were ±3.9% and ±7.3%within-day and between-day respectively. The lowest detectable concentration was 0.28 μg/mL.Conclusion The method is rapid,accurate and reproducible,which can provide bases for the study of its pharmacokinetics.
