摘要
目的建立鹿角脱盘蛋白提取工艺。方法采用SDS-聚丙烯酰胺凝胶电泳法(SDS-PAGE),考察不同pH值、不同料液比、不同浸提时间、不同盐浓度等影响因素对蛋白提取效果的影响。结果鹿角脱盘蛋白的提取工艺为:pH值12的缓冲液(含50mMEDTA、0.5MNaCl)浸提,料液比为1∶5,浸提时间为3h,浸提次数2次。结论验证实验结果表明,鹿角脱盘总蛋白收率可达2%,纯度可达80%。优选出的工艺稳定、合理、可行。
Objective To establish the extraction process of proteins from Sika antler base.Methods SDS-polyacrylamide gel electrophoresis(SDS-PAGE),was used to study the impacts on protein extraction,including pH values,solid-liquid ratio,extraction times,salt concentration and other factors.Results The extraction process was obtained as follows:the materials were extracted twice with pH12 buffer solution in which contained 50 mmol/L EDTA and 0.5 mol/L NaCl,each time adding 5 times amount of buffer solution and extracting 3 h.Conclusion The yield of proteins can be up to 2%,and purity can also be up to 80%.The optimum process obtained is steady,reasonable and feasible.
