肝片吸虫虫体抗原氨基酸成分分析

摘要 对来自水牛、黄牛体内的肝片吸虫虫体抗原的氨基酸组份进行研究,并对黄牛体内虫体进行先超声裂解后冷浸或先冷浸后超声裂解处理以比较其抗原氨基酸组份的变化。结果表明先超声裂解后冷浸的脱脂和未脱脂的水牛、黄牛虫体抗原氨基酸含量分别为374.035mg/dl、396.318mg/dl和119.58mg/dl、243.341mg/dl,水牛虫体的氨基酸明显高于黄牛虫体的氨基酸。而先冷浸后超声裂解处理的脱脂和未脱脂的黄牛虫体抗原氨基酸含量分别为101.352mg/dl和169.982mg/dl,不同处理方法对黄牛体内氨基酸组份影响较大,先超声裂解后冷浸优于先冷浸后裂解,提示制备肝片吸虫虫体抗原最好以水牛虫体进行先超声裂解后冷浸,这样可得到高含量。 The antigen amino acids content of F.hepatica in water buffalo and cattle were studied and the changes of amino acids content of F. hepatica in cattle were investigated in this article.The results showed that the amino acids contents of de-fat and unde-fat antigens of F.hepatica in water buffalo and cattle were 374.053mg/dl,396.31mg/dl,119.958mg/dl and 243.341mg/dl,respectively.The amino acids content of F.hepatica in water buffalo is higher than that in cattle.But first cold soakage then ultrasonic decomposition de-fat and unde-fat antigen amino acids content in cattle with different are 101.325mg/dl and 169.982mg/dl respectively.There is great effect on the amino acids of F.hepatica in cattle with different treatment methods.First ultrasonic decomposition then cold soakage is better than first cold soakage then ultrasonic decomposition.It showed that it is the best to make antigen of F. hepatica in water buffalo with ultrasonic decomposition the cold soakage,which can obtain higher content and purity antigen.
