Determination of Acid Content in Clerodendranthus Spicatus (Thunb.) C.Y.Wu

目的:建立高效液相色谱法测定肾茶中主要有效成分之一熊果酸含量的方法,对提取条件和测定条件进行筛选,找出肾茶中熊果酸的含量测定的合适方法。方法:采用反相高效液相色谱法测定肾茶中熊果酸的含量。色谱条件为:流动相为乙腈-0.1%磷酸(78:22);色谱柱为Eclipse XDB-C18柱(150mm×4.6mm,5μm);紫外检测波长210nm;流速1.0mL/min;柱温35℃。结果:经测定熊果酸在1～6μg的范围内与峰面积呈现良好的线性关系,其回归方程为y=469.001676X+0.0690635,r=0.99999;平均回收率为98.76%,RSD=1.18%。结论:本方法简便、快速,结果准确、可靠,可作为肾茶药材中熊果酸的含量测定方法。

Objective:To establish an effective method for the determination of the acid content in Clerodendranthus spicatus by HPLC and select the extraction and determination conditions for the most appropriate method. Methods: RP-HPLC was used to determine the acid content in Clerodendranthus spicatus, with the mobile phase acetonitrile-0.1% phosphoric acid (78∶22), Eclipse XDB-C18 column (150mm×4.6 mm, 5μm), the UV detection wavelength at 210 nm, the flow rate 1.0 ml/min and the column temperature 35. Results: The determined acid showed a good linear relationship with the peak area at the range of 1-6 μg, with the regression equation, y=469.001676X+0.0690635, r=0.99999, and 98.7% of average recovery, RSD=1.18 %. Conclusion: The method is simple, quick, accurate and reliable and can be used to determine the acid content in Clerodendranthus spicatus.
