摘要
目的对血红蛋白(Hb)为基础的红细胞代用品以人脐带血Hb及其聚合Hb氧合曲线的测定方法进行研究。方法 Hb与氧结合的能力通常用氧分压[p(O2)]和Hb氧饱和度(Y)构成的氧合曲线来表示。制备人脐带血纯化Hb、聚合Hb,测定高铁Hb浓度,用高效液相色谱(HPLC)测定各种Hb浓度,做氧合曲线。结果改进检测方法和装置后,得到S形的氧合曲线,测得人脐带血纯化Hb样品的[p(O2)]50为(0.934±0.009)kPa[(7.17±0.07)mmHg],5′-磷酸吡哆醛(PLP)修饰的聚合Hb[p(O2)]50为(2.727±0.033)kPa[(20.50±0.25)mmHg]。结论该方法设备简单、操作方便、结果可靠、重复性和重现性较好。
Objective To study the determination method of hemoglobin (Hb) based red blood cell substitute by means of measuring human umbilical cord blood Hb and polymerized Hb oxygenation curve. Methods The binding capacity of Hb and oxygen were commonly expressed by oxygenation curve which constituted partial pressure of oxygen[p(O2)] and Hb oxygen saturation level(Y). The purified human cord blood Hb were prepared and polymerized, the methemoglobin concentration of Hb was determined and various concentration of Hb were determined by high performance liquid chromatography(HPLC). Then the oxygenation curves were drawn. Results With the improved detection methods and devices, S-shaped oxygen equilibrium curves was obtained and the [p(O2)]50 of the purified Hb of human umbilical cord blood was (0.934 ± 0.009) kPa [(7.17 ± 0.07) mmHg] and 5′-pyridoxal phosphate(PLP) modification of the polymerization of Hb was (2.727 ± 0.033) kPa[(20.50 ± 0.25) mmHg]. Conclusion It is demonstrated that this approach has the following advantages ie. simple, reliable, convenient, repeatable and reproducible.
