组织切片用胃粘膜固定方法的技术改进

Technical Improvements of Fixing Histological Section by Gastric Mucosa

运用常规固定方法、部分胃分离后去渣固定方法和切口去渣缝合后充盈固定方法,对兔、狗和猪的胃进行了固定。经过石蜡包埋切片,HE染色,光镜观察后将这几种固定胃组织的方法进行了比较。结果表明,用常规固定方法固定的胃,胃粘膜有很多地方存在破损,结构模糊;用部分胃分离后去渣固定方法和切口去渣缝合后充盈固定方法固定的胃,不仅胃粘膜完整,而且还保持了胃壁的原有形态,不仅适用于制作组织切片,而且也可以作为胃粘膜肉眼宏观观察的材料。

The conventional fixation method,the fixation method that removed the dregs after the stomach separated,and the fixation method that filled the stomach after cut the stomach and removed the dregs and then sewed the incision were used to fix stomachs of rabbit,dog and pig.Then these three fixed stomach tissues were embedded by paraffin,and the paraffin-embedded sections were stained with the HE-stain method and observed under the optical microscope.Comparing these methods,it found that the gastric mucosa which fixed by the conventional fixation method was damaged seriously;the stomach which fixed by the fixation method that removed the dregs after the stomach separated and the fixation method that filled the stomach after cut the stomach and removed the dregs and then sewed the incision not only could keep the mucosa completely,but also maintained its original shape.It not only fit for making histological sections,but also can be used as the materials for observation by eye.
