水通道蛋白4在大鼠外伤性脑损伤的表达及意义 被引量:1

摘要 通过建立大鼠外伤性脑损伤动物模型,采用IgG免疫组化染色法、干湿比重法测定脑组织含水量,Evans Blue(EB)测定法观察大鼠血-脑屏障(BBB)通透性变化,半定量逆转录聚合酶链反应(RT-PCR)法检测脑组织AQP4 mRNA表达及其变化,研究急性实验性大鼠外伤性颅脑损伤水通道蛋白4(AQP4)的表达规律、急性脑水肿产生的关系,探讨大鼠外伤性脑损伤导致急性脑水肿中AQP4的表达变化及其与血、脑脊液屏障(blood-brainbarrier,BBB)通透性的关系,分析AQP4在外伤性脑水肿的分子生物学机制。 Through the establishment of rat animal model of traumatic brain injury,using IgG immunohistochemistry staining,wet and dry weight determination of brain water content,Evans Blue(EB) Determination of rat blood-brain barrier(BBB) permeability changes in semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) to detect expression of AQP4 mRNA in brain tissue and its changes,study the acute experimental traumatic brain injury in rats with water-channel protein 4(AQP4) expression in the laws,acute generated by the relationship between brain edema,and explore the traumatic brain injury in rats lead to acute brain edema in AQP4 expression and its relationship with the blood brain barrier(blood-brainbarrier,BBB) permeability relationship,analyze thr Molecular biological mechanisms of AQP4 in traumatic brain edema.
