C5-siRNA对大鼠心肌缺血损伤的保护作用

摘要 目的:研究C5-siRNA对大鼠心肌缺血的保护作用。方法:30只SD大鼠随机分为假手术组,心肌缺血模型组和C5-siRNA干预组。结扎模型组和C5-siRNA组大鼠的冠状动脉(以下简称冠脉)左前降支近端制备急性大鼠心肌缺血模型,在冠脉左前降支支配的中心区分别注射0.9%氯化钠液和C5-siRNA100μL.kg-1,1h后松扎,测定血流动力学指标、心电图改变及血清乳酸脱氢酶(LDH)和肌酸激酶MB(CK-MB)变化,光镜下观察心肌形态学改变。结果:C5-siRNA对心肌缺血损伤大鼠血流动力学指标均有一定的改善趋势;能缩小大鼠结扎冠脉后心肌缺血范围(N-ST)及心肌缺血程度(ST段位移值),减轻心肌病理学损伤;对严重的心肌缺血引起的血清乳酸脱氢酶和肌酸激酶同功酶升高也有明显降低作用,尤其是其中CK-MB与模型组相比差异显著(1109±245比1513±335,P<0.05)。结论:C5-siRNA对大鼠急性心肌缺血具有保护作用。 Objective:To study the protective effect of C5-siRNA on myocardial ischemia-reperfusion injury in rats.Methods: Thirty healthy Wistar rats were randomly divided into Sham group,control group and C5-siRNA group.The rat models of control group and C5-siRNA group were established by ligating the left anterior descending coronary arteries(LADs).The rats were infused with 100 μl·kg-1 of C5-siRNA into vein in C5-siRNA group and equal quantity of normal sarine in isehemia-reperfusion(IR) group after ligating the LADs 20 min for 30 min and then performing repefusion for 120 min.The parameter of dynamics,electrocardiogram,lactate dehydrogenase(LDH) and creatine kinase MB(CK-MB) in serum were measured.Morphology of myocardial tissue was observed with optics microscope.Results: C5-siRNA may improve the parameter of dynamics,and reduce the degree of ECG N-ST and ST-segment in IR rats.Myocardial fiber necrosis and inflammatory cell infiltration were found in IR group,but there was no necrosis and relatively little inflammatory cell infiltration in C5-siRNA group.Compare with Control group,serum LDH and CK-MB in C5-siRNA group were diminished significantly(1109±245 vs 1513±335,P0.05).Conclusions: C5-siRNA could prevent myocardial ischemia-reperfusion injury in rats by reducing inflammatory cell infiltration.
