摘要
通过不同植物生长调节剂的组合和配比,对五鹤续断茎尖、幼嫩茎段和叶进行愈伤组织的诱导、丛生芽分化和增值及生根培养,确定了五鹤续断组织培养的最佳培养基为:(1)初代培养基为MS+6-BA1.5 mg/L+NAA0.1mg/L;(2)丛生芽培养基为MS+6-BA 2.0mg/L+NAA 0.6 mg/L;(3)生根培养基为1/2MS+IBA 0.4 mg/L+PP3330.4mg/L.
Shoot tip and tender stems which sprouted at the beginning of spring were used as explants to study effect of plant growth regulator on tissue culture of Wu-He Dipsacus asper.The optimal media were(1)callus medium,MS+6-BA 1.5 mg/L+NAA 0.2 mg/L;(2)clump bud induction medium,MS+6-BA 2.0(mg/L)+NAA 0.5(mg/L);(3)rooting medium,1/2MS+NAA(1.0 mg/L)+ PP333(0.4 mg/L).
