摘要 目的:研究从芡实中提取多糖的方法,并建立其含量测定方法。方法:考察了加热回流、不同温度浸提、超声波振荡等方法提取芡实多糖,采用苯酚-浓硫酸方法显色,利用紫外分光光度计在489nm处进行测定多糖含量。结果:芡实多糖于45℃超声波振荡提取效果最佳,在10-70μg.mL^-1的范围内,样品的浓度与吸光度有良好的线性关系,线性回归方程为A=12.071C-0.0123,r=0.9990。结论:超声波振荡提取多糖,方法简单,提取率高,解决了芡实中淀粉、蛋白等物质的干扰问题,其含量测定方法快速简便,灵敏度高,可用于芡实多糖的含量测定。 Objective:Study on the extraction methods of the polysaccharide content in Gordon nut,and establish a method for the quantitative determination.Methods:Inspect the extraction methods of the polysaccharide content in Gordon nut with heat reflux,water bath at the deferent temperature and ultrasonic extraction;With C6H5OH-H2SO4 coloration,in use spectrophotometric method was used to analyze polysaccharide content in Gordon nut,the detection wavelength was at 489 nm.Results:Ultrasonic extraction at 45℃ is the optimal method of the polysaccharide content in Gordon nut,The sample concentration was good linear relationship with absorbance within 10~70μg·mL^-1.And the equation of linear regression was A=12.071C-0.0123,r=0.9990.Conclusion:Ultrasonic extraction method of the polysaccharide content is simple,high extraction rate,solved interference problem of starch,protein and other material.The method for the quantitative determination is accurate,and sensitive,and can be used to determine the polysaccharide content in Gordon nut.
