通冠胶囊对大鼠颈动脉内皮损伤后血管VEGF eNOS表达和血清NO浓度的影响 被引量:6

Effects of Tongguan Capsule on the Expression of VEGF eNOS in Intima and Serum Level of NO in Rats after Carotid Endothelium Injury

摘要 目的:探讨通冠胶囊促进动脉损伤内皮修复的分子机制。方法:建立大鼠左颈总动脉内皮损伤模型,24只雄性SD大鼠随机等分为治疗组、对照组和模型组,治疗组给予通冠胶囊水煮液(600mg/kg)灌胃,对照组给予辛伐他汀(2mg/kg),模型组予生理盐水(10mL/kg);8周后,取材HE染色和弹力纤维染色,计算机图像分析系统检测颈动脉内皮损伤处内膜面积(intima area,IA)、中膜面积(media area,MA)和内膜/中膜面积比(IA/MA),免疫组织化学法检测VEGF和eNOS的表达,分光光度法测定血清NO水平。结果:治疗组、对照组的IA和IA/MA均小于模型组(P<0.01),治疗组、对照组血管内膜VEGF、eNOS表达和血清NO水平与均高于模型组(P<0.05或P<0.01),治疗组的血清NO水平高于对照组(P<0.05)。结论:通冠胶囊促进损伤内皮修复的分子机制可能与VEGF-AKT-eNOS-NO信号转导通路有关。 Objective:To investigate the molecular mechanism of Tongguan Capsule accelerating the recovery of arterial endothelium injury.Methods:24 male Sprague-Dawley rats which the left common carotid arteries injured were randomly divided into3 groups(n=18):Group 1(10mg·kg^-1·d^-1 of PS was given after injury),group 2(2mg·kg^-1·d^-1 of Simvastatin was given after injury)and group 3(600mg/kg/d of Tongguan capsule was given after injury).After 8 week,the injured left common carotid arteries were removed for HE and elastic fibers staining and intima area(IA),media area(MA)and IA/MA were analyzed by Computer Image Analyzer.The expression of VEGF and eNOS in injured endothelium was detected with immunohistochemistry.The serum level of NO was determined by pectrophotometry.Results:The morphological analysis demonstrated that both Tongguan Capsule and Simvastatin remarkably decreased the area of intima and IA/MA.In comparison with Group l,Tongguan Capsule up-regulated the expression of VEGF and eNOS in tunica intima after balloon injury and increased the serum level of NO.The expression of VEGF and eNOS of Group 2 were similar to Group 3.The serum level of NO in Group 3 was higher than the level of Group 2.Conclusion:The mechanism which Tongguan capsule could promote the recovery of injured endothelium probably involves the VEGF/AKT/eNOS/NO signaling pathway.
