摘要 细菌人工染色体(BAC)载体由于稳定复制、嵌合体少、分离纯化简单、转化效率高等优点,在构建基因组文库中得到了广泛的应用.BAC基因组文库构建的一般过程,主要有载体和高分子量DNA的制备、大片段DNA和载体的连接、重组载体转化受体感受态细胞、阳性克隆的挑取、文库质量的鉴定等.BAC基因组文库在基因克隆、基因组物理图谱的构建、基因组测序、比较基因组学的研究中有着十分广泛的应用.随着BAC载体的发展和更多生物BAC文库的构建,BAC基因组文库将会对基因组学研究产生重要的影响. Bacterial Artificial chromosome (BAC)vectors are broadly used in genomic library construction because of its advantages of stabilized replication,less chimera,easily isolating and purifying,high transformation rating,etc.Protocol for construction of BAC genomic library includes preparation and ligation of BAC vectors and HMW DNA,transformation of ligation mixture into competent cells,selecting of recombinant cloning,and characterization of BAC genomic library.BAC genomic library has been broadly used in gene cloning,genome physical mapping,genomic sequencing,and comparative genomic research.Genomics will be influenced immensely because of the developments of BAC vectors and the constructions of more BAC libraries.
