软骨多糖通过线粒体途径诱导L1210细胞凋亡 被引量:1

摘要 研究了软骨多糖诱导L1210细胞发生凋亡的具体机制。采用TUNEL检测细胞凋亡,化学发光法及免疫印迹法检测Caspase-8、Caspase-9及Caspase-3/7酶活性变化,细胞免疫荧光检测Bax/Bcl-2蛋白变化。结果表明软骨多糖可促进L1210细胞发生凋亡,提高Bax/Bcl-2的比率,活化了Caspase-9和Caspase-3蛋白酶,而Caspase-8蛋白酶活性无变化。这些结果表明软骨多糖诱导L1210细胞通过线粒体途径发生了凋亡。 The mechanism of promotion effect of cartilage polysaccharides (PS) on apoptosis of L1210 cells were studied in this paper. The apoptotic cell death was detected by TUNEL assay, and Chemiluminescence and Western blotting methods were used to detect the activity of Caspase-8, Caspase-9 or Caspase-3/7. Cellular immunofluorescene method was used to detect changes of Bax/Bcl-2. Results showed that PS led to obvious apoptotic of L1210 cells, improved the Bax/Bcl-2 ratio and activated Caspase-9 and Caspase-3. But it showed little effect on Caspase-8. This research indicated the activation of the mitochondrial apoptotic pathway in response to the treatment of L1210 cells with PS.
