九孔鲍鱼纤维素酶的分离纯化与性质分析 被引量:7

摘要 采用SP-Sepharose阳离子葡聚糖凝胶交换柱层析、Sephacryl S-200丙烯葡聚糖凝胶柱层析和Hydrox-yapatite羟基磷灰石柱层析,从九孔鲍鱼(Haliotis diversicolor)消化腺中分离纯化到一种纤维素酶。SDS-PAGE结果显示,该酶的分子质量约为55kDa。该酶水解羧甲基纤维素钠的最适温度50℃,最适pH5.5。该酶在温度40℃以下,pH5.0~7.0之间具有较好的稳定性。 A cellulase was isolated and purified from the digestive tract of abalone (Haliotis diversicolor) by column chromatographies of SP-Sepharose,Sephacryl S-200 HR,hydroxyapatite and phenyl sepharose FF prepacked column.The molecular mass of the enzyme was approximately 55 kDa as estimated by SDS-PAGE.Optimal temperature and pH were 50 ℃ and 5.5,respectively.The enzyme was stable up to 40 ℃ under the pH range of 5.0~7.0.
