多粘菌素B可消除内毒素对脂质体介导基因转染的影响

摘要 目的探索应用多粘菌素B(PMB)阻止内毒素(LPS)对转染影响。方法用活性检测法观察LPS及PMB对脂质体-DNA复合物转染培养细胞后基因表达的影响。结果质粒制剂中LPS的存在明显降低转染后基因表达水平;培养基中加用PMB(30μg/ml)可去除LPS的这种作用。结论转染过程中PMB的添用可清除LPS对转染的不利影响。 Objective: To study the prevention of the effects of endotoxin (or LPS) on gene transfection by polymyxin B (PMB). Methods: Using activity assay, the effects of endotoxin and PMB on gene expression levels were identified. Results: The existence of LPS in DNA preparation reduced the gene expression remarkably during gene expression while the preparation with PMB (30μg/ml) could get rid of the effect. Conclusion: The use of LPS-binding antibiotic PMB during transfection can eliminate the harmful effects of LPS on gene expression.
