摘要 目的研究凝血酶对正常人肺成纤维细胞(normalhuman lung fibroblasts,N-LF)和特发性肺纤维化(idiopathic pulmonary fibrosis,IPF)病人肺成纤维细胞(F-LF)单核细胞趋化蛋白-1(MCP-1)表达的影响。方法 N-LF和F-LF细胞分别培养于96孔培养板内,凝血酶诱导4和24h后,收集上清液并用ELISA方法检测MCP-1蛋白水平。结果凝血酶可诱导N-LF和F-LF细胞释放MCP-1,但F-LF细胞的MCP-1分泌水平远远高于N-LF细胞。结论人肺成纤维细胞在肺纤维化过程中过度合成和释放MCP-1,凝血酶可进一步促进其过度表达MCP-1。 Objective To investigate the effects of thrombin on monocyte chemoattractant protein-1 (MCP-1) protein release and mRNA expression in both normal human lung fibroblasts (N-LF) and fibroblasts derived from IPF (idiopathic pulmonary fibrosis patient) (F-LF). Methods N-LF and F-LF were cultured in a 96-well culture plate. Cells were stimulated with thrombin (10 nmol/L) for 4 and 24 h. Culture supernatants from 96- well plates were collected and MCP-1 protein levels in supernatants were assessed by ELISA. Results Thrombin can induce N-LF and F-LF MCP-1 release, and the MCP-1 levels in F-LF are much higher than the MCP-1 levels in N-LF. Conclusion Fibroblasts MCP-1 mRNA derived from patients with IPF overexpress MCP-1 and thrombin can induce its MCP-1 expression with high efficiency.
