RP-HPLC法测定醋青皮配方颗粒中橙皮苷的含量

Determination of Content of Hesperidin in Cuqingpi Dispensing Granule by RP-HPLC

目的:建立醋青皮配方颗粒中橙皮苷含量的测定方法。方法:运用正交试验优化样品处理的提取条件,采用高效液相色谱法检测橙皮苷含量,色谱柱为Shim-packVP-ODS柱(4.6mm×250mm,5μm);流动相为甲醇-0.1%磷酸(35∶65);流速1mL·min-1;检测波长284nm。结果:橙皮苷在0.1120～1.120μg呈良好的线性关系(r=0.9999),平均回收率为102.63%,RSD为1.24%。结论:该法简便可行、快速,可用于醋青皮配方颗粒的质量控制。

Objective：To establish a HPLC method for the determination of hesperidin in Cuqingpi Dispensing Granule.Method：After using the orthogonal test to optimize extracting conditions,hesperidin was separated on Shim-pack VP-ODS （4.6 mm×250 mm,5 um）column and detected at 284 nm,with methanol-0.1% phosphoric acid solution （35∶ 65） as amobile phase,and the flow rate was 1.0 mL·min-1.Result：Hesperidin was linear in the range of 0.112 0～1.120 μg （r=0.999 9） .The average recovery of hesperidin was 102.63% with RSD 1.24% .Conclusion：The method is simple and rapid which can be used for the quality control of Cuqingpi Dispensing Granule.
