摘要
目的:对川芎中阿魏酸效应组分进行提取、纯化,研究阿魏酸效应组分对酪氨酸酶单酚酶活力的抑制效应。方法:以阿魏酸为主要指标,采用乙醇提取,大孔树脂吸附法的纯化工艺,用HPLC进行含量测定,采用酶动力学方法,分析不同浓度阿魏酸效应物对酪氨酸酶的抑制效应。结果:提取物中阿魏酸效应物得率为2.8%,效应物中阿魏酸平均含量为11.2%,以阿魏酸计其IC50为0.0048g.L-1,能使酪氨酸酶反应的迟滞时间明显延长。结论:提取纯化方法简便可行,本实验可为开发中药美白产品提供依据。
OBJECTIVE To study on extraction and purification of ferulic acid from Ligusticum wallichii Franch on and the inhibitory effect of active parts on tyrosinase. METHODS Extraction process was adding alcohol in Ligusticum wallichii Franch on, purification process was using macro porous resins, ferulic acid as marker was monitored by HPLC,inhibitory effect of ferulic acid on tyrosinase was analyzed of the enzyme kinetics with different concentration. RESULTS Extractive rate was 2. 75%, the content of ferulic acid in extraction was 11.24%, IC50 was 0. 004 8 g·L^-1, time lag of reaction was extended. CONCLUSION The method is simple and feasible, the active parts of ferulic acid from Ligusticum wallichii Franeh on have strong whitening efficacy.
