三种中药皂甙对K_(562)细胞诱导分化作用的实验研究

EFFECT OF TOTAL SAPONINS OF THREE CHINESE TRADITIONAL MEDICINES ON INDUCED DIFFERENTIATION OF K 562 CELLS

目的研究人参总皂甙（ＴＳＰＧ）、绞股蓝总皂甙（ＧＰ）、三七总皂甙（ＰＮＳ）对Ｋ５６２细胞诱导分化的影响；方法采用细胞体外培养技术，通过细胞形态学、细胞化学、表面膜抗原（ＨＩＲ２、ＣＤ１５、ＣＤＷ４１）等检测方法；结果经三种皂甙诱导后，ＴＳＰＧ、ＧＰ不能诱导Ｋ５６２细胞合成过氧化物酶（ＰＯＸ）而能合成血红蛋白（Ｈｂ），仅有表达ＣＤＷ４１，ＨＩＲ２的阳性细胞数明显增加；而ＰＮＳ不能诱导Ｋ５６２细胞合成Ｈｂ而能合成ＰＯＸ，表达ＣＤＷ４１、ＨＩＲ２、ＣＤ１５的阳性细胞数均明显增加；结论ＴＳＰＧ、ＧＰ、ＰＮＳ均能诱导Ｋ５６２细胞向多系分化为较成熟的细胞，ＴＳＰＧ、ＧＰ主要诱导其向红系分化。

Objective The effect of total saponins of panax ginseng(TSPG).Gynostemma pentaphllun (GP) and panax notoginseng (PNS) on the induced differentiation of K 562 cells were observed. Methods We used cells culture ex vivo,morpholgical observation. cytochemistry and analysis of Smcd antigth. Results TSPG.GP couldn't induce K 562 cells synthesize peroxisome (POX) but PNS could, on the other hand,PNS couldn't induce K 562 cells synthrsize hemoglobin(Hb) but TSPG.GP could.In addition CD W41 、HIR 2、CD 15 positive cells were significantly increased after PNS induction, While HIR 2 CD W41 positive cells were increased after TSPG GP induction Conclusion TSPG GP PNS could induce K 562 cells towards erythrocytic lineage generally, while PNS might induce its towards granulocytic lineage mainly.
