摘要 目的研究肿瘤坏死因子(TNF)α多克隆抗体对新生大鼠缺氧缺血性脑损伤(HIBD)后大脑的保护效应。方法制备左脑HIBD的新生大鼠模型,于HIBD前后分别腹腔注射一定浓度的TNFα多克隆抗体,观察用药前后损伤大脑脑水含量及组织病理学变化。结果HIBD前后腹腔注射TNFα多克隆抗体可明显降低脑水含量(P<0.01),大脑水肿明显减轻,神经元死亡明显减少,但对其相应mRNA转录无影响。结论TNFα多克隆抗体可以拮抗TNFα的炎性作用。 Objective To study the protective effects of polyclonal antibody to tumor necrosis factor(TNF)α on hypoxic ischemic brain damage (HIBD) of neonatal rats. MethodsWe made a model of left brain HIBD in neonatal rats and injected TNFα polyclonal antibody into abdominal cavity before and after HIBD. Brain water content and histopathology were observed. Results Polyclonal antibody of TNFα administered before or after HIBD decreased brain water content significantly ( P <0.01), but had no effects on corresponding mRNA expression. TNFα polyclonal antibody also attenuated edema of brain and death of neurons pathologically. Conclusions TNFα polyclonal antibody may antagonize the inflammatory function of TNFα and protect brain after HIBD.
