摘要
以16种基因型的二倍体甜菜(Beta vulgaris L.)为材料,建立并优化了甜菜的植株再生体系;对影响甜菜转基因的相关因素进行了比较分析,确定了抗生素标记基因的适宜筛选浓度为200mg/L;菌液适宜浸染浓度OD600为0.3;浸染最适时间为10min;外植体与农杆菌适宜共培养时间为4d;添加乙酰丁香酮的适宜浓度为100μmol/L。研究结果的获得,为完善甜菜的遗传工程以及甜菜基因组学的相关研究奠定基础。
Sugarbeet (Beta vulgaris L.) varieties including 16 kinds genotype were chosen, its plant system was established and optimized, and related factors on genetic transformation were compared an generation d analyzed as well. In this study, optimal dose of Kanamycin 200 mg/L was identified; optimal infected Agrobacterium tumefaciens EHA105-OsAPX3 was OD600 = 0.3; and the optimal infected internal was 10min; the optimal explants co-cultured with agrobacterium was 4 days; the optimal concentration of acetosyringone added was 100μmol/L. The results would make full usage of genetic transformation and genome on sugarbeet in the future.
