摘要 以黄花槐花粉为试材,利用I2-KI染色法、TTC(氯化三苯基四氮唑)染色法、培养基萌发试验法对花粉生活力进行测定,并研究了不同浓度的Ca2+对花粉萌发和花粉管伸长的影响。结果表明,I2-KI染色法、TTC染色法均不适合测定黄花槐花粉生活力;不同浓度的蔗糖和硼酸对花粉的萌发有促进作用,蔗糖20mg/L+硼酸2.0mg/L培养基花粉的萌发达30.2%;适当浓度的Ca2+能促进花粉萌发和花粉管的伸长,当Ca2+浓度为1.6×10-4mg/L时,花粉的萌发率最高,花粉管伸长最快。 The experiment used the Sophora x anthantha pollen as experimental materials, the pollen viability of Sophora x anthanthawas detected by methods of I2-KI, TFC dyeing and germination in vitro, to study the influence of the different concentration of the Ca2+on pollen germination and pollen tubes growth. The results showed that the method of I2-KI and TYC dyeing were not suitable to detect the pollen viability of Sophora x anthantha, different concentrations of sucrose and boric acid had great effects on the germination of pollen, when their concentration were 20 mg/L+2.0 mg/L, the germination rates of pollen reached 30.2%. The concentrations of Ca2+ could affected the spead of pollen germination and growth. When the concentration of Ca2+ was 1.6×10-4 mg/L, the pollen germination rate was the highest and growed quickly.
