去甲斑蝥素对大鼠糖尿病视网膜病变中VEGF表达的影响 被引量:3

Effect of Norcantharidin on the Expression of Vascular Endothelial Growth Factors in Diabetic Retinopathy Rats

摘要 目的观察去甲斑蝥素对大鼠糖尿病视网膜病变中血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响,探讨去甲斑蝥素治疗抑制糖尿病视网膜病变的可能机制。方法将40只SD大鼠采用1%链脲佐菌素(STZ)溶液60 mg.kg-1腹腔注射,诱导大鼠糖尿病模型。40只大鼠全部成模,随即将40只糖尿病大鼠按随机数字表法分为2组:糖尿病组和去甲斑蝥素治疗组,每组20只。另取20只SD大鼠作为正常对照组。去甲斑蝥素治疗组采用去甲斑蝥素28 mg.kg-1腹腔注射,2次.周-1,共8周。糖尿病组和正常对照组给予相应容量生理盐水腹腔注射,2次.周-1,共8周。干预8周后,取各组大鼠眼球分别行病理检查及免疫组化ELISA法测定VEGF浓度。结果糖尿病组、去甲斑蝥素治疗组VEGF在大鼠视网膜组织中表达水平均高于正常对照组(P<0.05或P<0.01),去甲斑蝥素治疗组VEGF在大鼠视网膜组织中表达水平低于糖尿病组(P<0.05或P<0.01)。病理检查结果示:正常对照组大鼠视网膜组织中VEGF阳性细胞微量表达;糖尿病组、去甲斑蝥素治疗组大鼠视网膜组织中VEGF阳性细胞均大量表达,但去甲斑蝥素治疗组阳性反应程度较糖尿病组减弱。结论去甲斑蝥素明显抑制大鼠糖尿病视网膜组织中VEGF表达水平。提示去甲斑蝥素可能通过降低大鼠糖尿病视网膜组织中VEGF表达,而缓解其糖尿病视网膜病变进程。 Objective To observe the effects and possible mechanism of norcantharidin the expression of vascular endothelial growth factors(VEGF) in the retinopathy of diabetic rats.Methods 40 SD rats were induced into diabetic models by 1% streptozotocin(60 mg·kg^-1) solution intraperitoneal injection,40 diabetic models were formed.The 40 diabetic rats were randomly divided into 2 groups by number table method:diabetes group and norcantharidin treatment group,20 rats in each group.Another 20 normal rats were recruited as normal control group.The norcantharidin treatment group was injected norcantharidin(28 mg·kg-1) while diabetes groups and normal control group were injected the corresponding capacity of saline solution into peritoneal cavity twice every week.8 weeks later,each group were removed the eyes and the expression of VEGF in the retinas were detected by pathological examination and immunohisochemistry ELISA.Results The expression of VEGF in retinas in diabetes group and norcantharidin treatment group was significantly higher than that of normal control group(P〈0.05 or P〈0.01),the expression of VEGF in norcantharidin treatment group was lower than diabetes group(P〈0.05 or P〈0.01).Pathological examination showed that the VEGF positive cells expressed slightly in retinas in normal control group,the diabetes group and norcantharidin treatment group expressed abundantly,and the VEGF positive cells in norcantharidin treatment group was lower than in diabetes group.Conclusion Norcantharidin injected intraperitoneally decreases the expression of VEGF in retinas of diabetic rats so that partly suppresses the development of diabetic retinopathy.
