摘要
采用有机介质中微生物转化的方法,对甾体C1,2位的生物脱氢进行了研究.通过考察有机溶剂对甾体底物和产物的溶解度,微生物细胞在不同有机溶剂中的存活率以及有机溶剂的加入量,构建了较合适的二甲苯/发酵液两相体系,且二甲苯加入量占总体积的15%为宜.然后以雄甾-4-烯-6-亚甲基-3,17-二酮为底物,利用简单节杆菌(Arthrobacter simplex)完整细胞在两相体系中对甾体A环C1,2位进行生物脱氢,得到抗肿瘤药物依西美坦(雄甾-1,4-二烯-6-亚甲基-3,17-二酮).对转化时间、转化温度、摇床转速、摇瓶装液量等条件进行了一系列的研究.结果表明,在装液量为20 mL每100 mL三角瓶,温度为32℃,转速为180 r/min,转化时间为24 h的条件下,转化率可达到13.6%.
Microbial transformation of steroid in organic medium was studied in this paper.The solubility of substrate and conversion production,the livability of microbial cells in organic solvent and the amount of added organic solvent were examined.Then an xylene/ fermentation broth two-phase system had been constructed.The xylene in the total volume of 15% was more appropriate.Using 6-methylene-androsta-4-ene-3,17-dione as substrate,which could be C1,2-biodehydrogenated by Arthrobacter simplex in the two-phase system,the antitumor drug—exemestane(6-methylene-androsta-1,4-diene-3,17-dione) was obtained.The conversion time,conversion temperature,shaking speed and bottled fluid volume had been discussed,respectively.All the results show that the conversion rate come to 13.6% with medium in volume of 20 mL per 100 mL flask,temperature in 32 ℃,speed at 180 r/min,and conversion time of 24 h.
