摘要
目的探讨皮下接种成瘤与原位接种成瘤在内环境不同情况下对肿瘤生长、侵袭转移能力的影响。方法选取人肝癌细胞系HepG2进行体外培养。将BALB/c裸鼠随机分为皮下瘤接种、肝脏原位细胞接种两组。观察裸鼠成瘤率、裸鼠存活率、肿瘤生长速度、肿瘤病理学特点及远处脏器转移。结果 HepG2细胞体外生长状态良好,增殖活跃。皮下接种成瘤率100%,肿瘤呈结节状膨胀性生长,对周围组织有侵袭,但无转移。肝脏原位细胞接种组成瘤率100%,肿瘤呈结节状膨胀性生长,对周围肝组织和腹壁有侵袭,肝内转移率70%,肺转移50%。结论 HepG2细胞皮下和肝原位接种易成瘤,可用于肝细胞癌(hepatocellular carcinoma,HCC)体内研究的实验模型。如果进行HCC的侵袭转移的机制及药物干预治疗等方面的研究,肝原位接种成瘤更能模拟人类肿瘤生长的条件。
Objective To detect the difference of subcutaneous injection and liver injection in tumor growth and metastasis.Methods HepG2 cells,human hepatoma cell line were cultured and injected into subcutaneous flank and liver lobe in mice randomly.The tumor formation rate,mice survival rate,tumor growth velocity,oncopathologie character and metastasis were detected.Results HepG2 cells grew well and proliferated actively in vitro.All of the subcutaneous injections developed tumor.The tumor grew fast and presented expansive growth pattern with surrounding invasion but had no metastasis.All the liver injection formed tumor with surrounding invasion,and intra hepatic and lung metastasis in 70% and 50% respectively.Conclusion Subcutaneous injection and liver injection of HepG2 cells can form tumor.This method can be used in animal model study in vivo.Liver injection experienced tumor development procedure more likely as human hepatocellular carcinoma(HCC).It is more useful in mechanism and treatment studies.
