摘要
目的筛选与涎腺腺样囊性癌转移相关的钙黏素(CDH)家族基因。方法对涎腺腺样囊性癌转移能力不同的两个细胞株的基因表达谱进行生物信息学分析,筛选出可能与转移相关的CDH家族基因,通过荧光实时定量PCR技术对相关基因的表达差异进行验证,并用免疫组织化学技术研究有差异的CDH基因在临床样本中的表达及其与转移的关系。结果生物信息学分析发现,表达谱中有14个CDH家族基因在两个细胞株中的表达有差异,实时定量PCR验证发现CDH4、CDH5、CDH12在两细胞株之间的表达差别有统计学意义(P<0.05),免疫组织化学发现CDH12在有转移的临床标本中的表达高于没有转移的标本((P<0.05)。结论 CDH12可能与涎腺腺样囊性癌的高转移能力有关。
Objective To explore the relationship of the expression of cadherin family genes and the metastasis of adenoid cystic carcinoma.Methods The genes of adenoid cystic carcinoma were screened through bioinformatics analysis from ACC-2 and ACC-M gene expression spectra.The correlative genes involving CDH family were determined.Real time-PCR was used to examine the mRNA level of these genes in the ACC-2 and ACC-M.Then immunohistochemical methods were used to investigate the expression of the genes which were found out above in 80 cases of adenoid cystic carcinoma.Results Forteen kinds of genes were detected from gene expression profile.Real time-PCR demonstrated that the expression of CDH4 and CDH12 in ACC-M were significantly higher as compared with ACC-2(P0.05).The expression of CDH5 in ACC-M were significantly lower as compared with ACC-2(P0.05).The expression of CDH12 was significantly higher in the tissues with metastasis and recurrence than without(P0.05).Conclusion CDH12 may play an important role in the metastasis of adenoid cystic carcinoma.
