瘦素在人溃疡性结肠炎中的表达及作用机制研究

The expression of leptin in human ulcerative colitis and its role in pathogenesis

目的观察瘦素在人溃疡性结肠炎组织中的表达及其作用机制。方法收集我院2006年6月~2009年6月确诊的溃疡性结肠炎患者35例,以结肠镜检查无明显异常者25例作为阴性对照。酶联免疫法观察血清细胞因子（TNF-α和IL-2）浓度的变化,免疫组化方法检测溃疡性结肠炎病理组织中瘦素的表达情况。结果在溃疡性结肠炎患者的病理组织中,瘦素的表达明显高于对照者,其血清中TNF-α的含量也高于对照者,IL-2则相反。结论瘦素在溃疡性结肠炎组织中过量表达,可能通过细胞因子TNF-α的上调和IL-2的下调诱导溃疡性结肠炎的发生。

Objective To observe the expression of leptin in human ulcerative colitis and to analyze the mechanism.Methods Thirty-five patients with ulcerative colitis diagnosed in our hospital from June 2006 to June 2009 were selected,25 normal colonoscopy people were selected as negtive controls.The concentrations of TNF-α and IL-2 in serum were detected by ELISA method in patients with ulcerative colitis,the expression of leptin was analyzed by immunohistochemical staining methods.Results The expression of leptin in patients with ulcerative colitis was higher than that in controls.The TNF-α level in blood was also higher than that in controls,but the IL-2 level had the opposite result.Conclusion The excessive expresion of leptin may cause ulcerative colitis through the up-regulation of TNF-α and the down-regulation of IL-2.
