摘要
目的 研究高压氧(HBO)预处理诱导脊髓神经元氧化耐受的机制.方法 取胚胎小鼠脊髓行原代神经元培养,分为正常对照组和HBO预处理组(HBO组).利用Westem Blot、RT-PCR等方法,观察HBO预处理对脊髓神经元抗氧化酶水平的影响.结果 HBO预处理前后超氧化物歧化酶(SOD)表达无统计学差异,血红素氧合酶(HO-1)在处理后表达增多.行RT-PCR检测HBO处理前后HO-1 mRAN水平,发现在预处理后4 h开始明显增多,处理后8 h表达达到高峰,直至处理后24 h其表达量仍比预处理前高.结论 HBO预处理对脊髓神经元氧化损伤的保护作用可能与诱导HO-1表达有关.
Objective To investigate mechanism of hyperbaric oxygen (HBO) preconditioninginduced tolerance to oxidative injury in primary cultured spinal cord neurons.Methods Spinal cord was obtained from fetal mice.Primary cultured neurons were divided into two groups:the control group and the HBO preconditioning group.Western Blot and reverse transcriptase polymerase chain reaction (RT-PCR) were used to observe levels of antioxidant enzyme in primary cultured spinal cord neurons.Results obtained from Western Blot showed that the level of antioxidant enzyme,heme oxygenase-1 (HO-1) in spinal cord neurons increased evidently after a single HBO exposure,reached peak at hour 8,and its expression 24 hours after HBO exposure was still higher than that before preconditioning.However,no significant differences could be noted in the expression of SOD between groups with or without HBO treatments.Conclusions The protective effect of HBO on oxidative injury in primary cultured spinal cord neurons might be involved in the expression of HO-1.
