摘要
目的:研究不同强度跑台训练大鼠肝铁调素调节蛋白(hemojuvelin,HJV)及转铁蛋白受体2(transferrin receptor2,Tfr2)mRNA的变化,探讨HJV和Tfr2在运动性低血红蛋白发生中的作用。方法:18只雄性Wistar大鼠(300±10g)随机分为对照组(CG)、适度运动组(MG)和过度运动组(SG)。对照组大鼠不运动,适度运动组和过度运动组大鼠分别进行不同负荷的递增负荷跑台训练。于运动5周后安静时取材,采用实时定量荧光PCR(Real Time RT-PCR)检测大鼠肝组织匀浆中HJV、neogenin(HJV受体)和Tfr2 mRNA表达;Western Blot检测肝细胞Tfr2蛋白表达。结果:(1)过度运动组大鼠HJV、neogenin和Tfr2 mRNA均显著高于对照组和适度运动组(P<0.01,P<0.05);(2)适度运动组大鼠肝Tfr2 mRNA和蛋白表达量显著低于对照组(P<0.05),HJV和neogenin mRNA表达量与对照组相比无显著差异(P>0.05)。结论:长时间大强度运动后肝HJV和Tfr2表达增加,这可能与运动性低血红蛋白的发生有关。
Objective The present study investigated the changes in hemojuvelin(HJV)and transferrin receptor 2(Tfr2)mRNA in rats after treadmill running with different loads for illuminating the mechanism of iron metabolism in exercise-induced hypohemoglobin.Methods Eighteen male Wistar rats were randomly divided into three groups:control group(CG),moderate exercise group(MG),and strenuous exercise group(SG)according to the intensity of running.The expression of HJV,neogenin(HJV receptor)and transferrin recaptor 2(Tfr2)mRNA in rats'liver was observed by real-time RT-PCR.The expression of hepatic Tfr2 protein was detected by Western Blot.Results(1)The expression of HJV,neogenin and Tfr2 mRNA in SG increased more significantly than those in both CG and MG(P 0.01)(.2) The expression of Tfr2 in MG significantly decreased as compared with that in CG(P 0.05),whereas there were no significant difference in HJV and neogenin mRNA between MG and CG.Conclusion The increased expression of hepatic HJV and Tfr2 after strenuous exercise is probably related to exercise-induced hypohemoglobin.
