RGD标记纳米氧化铁的肿瘤血管生成分子影像学研究 被引量:7

摘要 在肿瘤血管生成过程中,活化血管内皮细胞整合素表达明显上调.精氨酸-甘氨酸-天冬氨酸(arginine-glycine-aspartic acid,RGD)三肽序列是整合素ανβ3的特异性识别位点.本项研究制备了RGD肽标记的以聚乳酸(poly lactic acid,PLA)为包被材料的超小超顺磁性氧化铁(ultrasmall paramagnetic iron oxide,USPIO)(以RGD-PLA-USPIO指代),并在体外和体内实验中考察了其检测肿瘤血管生成的能力.普鲁士蓝染色和透射电子显微镜(transmission electronic microscopy,TEM)证实了人脐静脉血管内皮细胞(human umbilical vein endothelial cells,HUVECs)可以特异性摄取RGD-PLA-USPIO.Vx-2肿瘤模型注射RGD-PLA-USPIO后在磁共振T2加权图像可以发现显著的信号降低,而对侧肌肉组织仅见轻微本底改变.结果表明,RGD-PLA-USPIO可以有效在磁共振图像上显示活化肿瘤血管内皮整合素ανβ3的表达,未来有望在肿瘤的诊断和疗效监测方面发挥重要作用.
