高胆固醇饲料对豚鼠胆囊组织分泌糖蛋白的影响

Effects of high cholesterol diet on glycoprotein secretion from the gall bladder in guinea pigs

目的：应用组织培养方法，了解高胆固醇饲料对豚鼠胆囊上皮细胞合成分泌糖蛋白（ＧＰ）的影响。方法：豚鼠４０只，分为普通饲料组（Ａ组）与高胆固醇饲料组（Ｂ组），饲养１周。培养胆囊组织，以３Ｈ－氨基葡萄糖为底物，测定其合成分泌ＧＰ的功能；并加入氢化考的松和腹腔巨噬细胞（Ｍψ），观测对胆囊组织合成分泌ＧＰ的影响。结果：①Ｂ组的腹腔Ｍψ活力较Ａ组明显升高。②培养Ｂ组胆囊组织，释放入培养液中的ＧＰ含量比Ａ组显著增加。③在Ｂ组胆囊组织培养液中加入氢化考的松，胆囊组织释放ＧＰ明显受抑。④在Ａ组胆囊组织培养液中，加入“普Ｍψ”，胆囊组织释放ＧＰ的水平与对照水平接近。加入“高Ｍψ”，浓度较高时胆囊组织分泌ＧＰ增强。结论：①高胆固醇饲料可致豚鼠胆囊组织合成分泌ＧＰ功能增强。②高胆固醇饲料能增强豚鼠Ｍψ活力，可促进胆囊组织合成分泌ＧＰ。结合动物胆囊结石模型实验的结果推测，可能与豚鼠Ｍψ因高胆醇饲料活化，释放细胞因子ＴＮＦ、ＩＬ－１等有关。

Objective: To investigate the effects of high cholesterol diet on glycoprotein (GP) secretion from the gall bladder with tissue culture in guinea pigs. Methods: Forty guinea pigs were randomized into two groups, A and B. The animals in group A were fed with ordinary food and those in group B with that containing 1.2% cholesterol for one week. Then the gall bladder tissue was extracted and cultured. GP secretion from the gall bladder was observed using 3H glucosamine as a precursor. Meanwhile, Mψ was added to the culture medium to investigate its effects on GP secretion from the gall bladder. Results: The activity of peritoneum Mψ and the release of 3H glucosamine labeled GP were significantly increased in group B as compared with those in group A. Sixteen hours after Mψ was added to the culture medium, mucin secretion had an evident increase in group B. Hydrocortisone caused a reversible dose dependent inhibition of GP secretion from the gall bladder in group B and also inhibited the stimulatory effects of Mψ on GP secretion in group A. Conclusion: ① High cholesterol diet can induce the increase of GP secretion from the gall bladder in guinea pigs. ② Mψ can be activated by high cholesterol diet. Combining with the results of the studies on animal model of gall bladder stone, it is hypothesized that the increase in GP secretion from gall bladder might be related to the release of cytokines such as TNF, IL 1 etc. from Mψ activated by high cholesterol diet.
