摘要
淋球菌分型研究包括表型分型和基因分型研究。淋球菌多抗原序列分型方法是一种基于测序的、应用前景广阔的基因分型方法,其检测标本不仅可以是菌培养物,还可以直接用于拭子标本的分型研究。PCR分别扩增两个编码淋球菌外膜蛋白基因(por和tbpB)的高变区,PCR扩增产物测序后与淋球菌多抗原序列分型方法网站比对以获取菌株的基因型。淋球菌多抗原序列分型方法具有操作简单、结果客观、国内外实验室间结果可比性等优势,该方法已经在预测淋球菌耐药性、分析性网络、鉴定特殊菌株爆发和流行等方面广泛应用。
The typing of gonococcus involves phenotypic analysis and molecular genotyping. Neisseria gonorrhoeae multi-antigen sequence typing (NG-MAST) is a sequence-based genotyping method with a broad prospect. NG-MAST system is not only suitable for cultured specimens, but also for swab samples. It allows the typing of N.gonorrhoeae strains by blasting against databases at NG-MAST Web site following amplification of variable internal regions of por and tbpB genes of gonococcal strains using PCR and sequencing of PCR products. Compared with other genotyping methods, NG-MAST appears simple and objective, and tends to produce results with satisfactory comparability between national and international laboratories. This system has been extensively used in predicting gonococcal resistance, analyzing sexual networks, identifying outbreak and epidemic strains, and so on.
