摘要
根皮苷是苹果中主要的水溶性多酚化合物,对人体具有多种生理保健功能。为了获得高纯度的根皮苷,运用高效离心分配色谱技术对苹果树枝乙醇提取物中的根皮苷进行分离纯化。对两相溶剂体系、转子转速和流动相流速等参数进行系统的优化实验。结果表明,最佳两相溶剂系统为正己烷-乙酸乙酯-甲醇-水(1∶5∶1∶5,v/v),上相有机相为固定相,下相水相为流动相,转子转速为1500r·min-1,流速为3.0mL·min-1,紫外检测波长为287nm。在此条件下,在30min内,从纯度为17.91%苹果树枝乙醇提取物中一步分离纯化得到根皮苷,高效液相色谱法测定其纯度为91.20%,根皮苷的回收率为98.10%。
Phloridzin is a main water-soluble polyphenol in apple, and has various physiological healthy functions to human body. In order to obtain higher purity phloridzin, high performance centrifugal partition chromatography was applied to separate and purify phloridzin from the ethanol extract of apple branches. The satisfactory separation effect was obtained by optimizing the parameters such as the two-phase solvent system, rotation speed and flow rate. The optimal purification conditions of phloridzin could be concluded as follows: a two-phase solvent system of n-hexane-ethyl acetate-methanol-water (1:5:1:5, v/v), with the upper organic phase as stationary phase and the lower aqueous phase as the mobile phase; the rotation speed of 1 500 r·min-1, the flow rate of 3.0 mL·min-1 and the detection wavelength of 287 nm.Under these conditions,phloridzin was successfully purified from the ethanol extract of apple branches by one step in 30 min, with the increase of average concentration from 17.91% to 91.20%, as determined by HPLC, and the average recovery of phloridzin was 98.10%.
