摘要
目的进一步探讨青蒿琥酯对肺纤维化的治疗作用及机制。方法取对数生长期的人胚肺成纤维细胞(HELF)系HFL-Ⅰ细胞,随机分为观察1~3组及对照组,观察1~3组分别加入质量浓度为1、10、100 mg/L的青蒿琥酯,对照组加入等体积培养液继续培养。用天狼猩红染色法检测胶原蛋白的分泌水平,RT-PCR法测定Ⅰ型和Ⅲ型前胶原蛋白mRNA的表达。结果观察1~3组胶原表达量、Ⅰ型、Ⅲ型前胶原蛋白mRNA表达均显著低于对照组,且各观察组间两两比较有显著差异(P均<0.05)。结论青蒿琥酯具有抗纤维化的作用,可在一定程度上降低胶原分泌,其机制可能为下调Ⅰ型和Ⅲ型前胶原蛋白mRNA表达。
Objective To investigate the antifibrotic action and mechanism of artesunate furtherly.Methods Human embryonic lung fibroblasts(HELF) cell lines HFL-I were cultured to log phase in vitro and divided into 3 observed groups and control group,the former 3 groups were treated with artesunate at the mass concentration of 1,10,100 mg/L,the control group were given equivalent DMEM/F-12.The collegan synthesis was determined by sirius red staining,the expression of type Ⅰ procollagen and type Ⅲ procollagen mRNA was detected by RT-PCR.Results Compared with the control group,the expression of collagen and type Ⅰ procollagen,type Ⅲ procollagen mRNA in observed groups decreased significantly,and remarkable differences were also seen among observed groups(P0.05).Conclusion Artesunate can resist pulmonary fibrosis and reduce the secretion of collagen in some extent,the mechanism may relate to the reduction of the expression of type Ⅰ procollagen and type Ⅲ procollagen mRNA.
