摘要
目的:研究p16,CyclinD1在胃癌发生过程中的表达。方法:采用免疫组化ABC法,检测胃癌、不典型增生、萎缩性胃炎及正常胃粘膜组织p16和CyclinD1的表达。结果:p16蛋白在正常胃粘膜表达率最高(867%,13/15),并随病变进展(浅表性胃炎-萎缩性胃炎-不典型增生-腺癌),表达率呈下降趋势(分别为85%;786%;318%;483%),其中不典型增生、腺癌组与正常组比较差异显著(P<005)。CyclinD1过度表达在萎缩性胃炎即已出现,并随病变发展表达率升高,萎缩性胃炎、不典型增生、腺癌的CyclinD1蛋白表达率分别为179%(5/28);364%(8/22);534%(31/58),其中腺癌与萎缩性胃炎差异显著(P<005)。相关性分析显示,大多数受检胃癌组织中,p16与CyclinD1呈反向表达。结论:p16和CyclinD1在胃上皮癌变过程中起着重要作用,其在胃癌中的反向表达趋势提示两者可能存在相互抑制机制。
p16 and Cyclin D1 expressions were
determined in the specimens of gastric carcinoma, atypical hyperplasia, atrophic gastritis,
superficial gastritis and normal gastric mucosa by using immunohistochemical method
ABC).The results showed that positive immunostaining rate for p16 protein was the highest in
normal gastric mucosa and decreased with progression from superficial gastritis to atrophic
gastritis to atypical hyperplasia and to adenocarcinoma (85%, 78.6%, 31.8%,
48.3%respectively). Positive immunostaining of cyclin D1 could be observed in atrophic
gastritis with the lesions progressing from atrophic gastitis to atypial hyperplasia and to
adenocarcinoma, the expression rates increased in order 17.9%, 36.4%, 53.4%respectively.
There was a significant difference between adenocarcinoma and atrophic gastritis group (P<
0.05). An interesting observation was that inverse expression between p16 and Cyclin D1 was
shown in most of the gastric cancer tested. The results suggest that p16 and Cyclin D1 play an
important role in gastric carcinogenesis, and that the inverse expression between p16 and
Cyclin D1 suggests antagonism between the two.
