摘要
目的动态观察大鼠实验性肝癌形成过程中抑凋谢基因bcl-2蛋白表达与γ-谷氨酰转肽酶(γ-GT)活性和谷胱甘肽硫转移酶(GST)活性改变的关系以及刺五加多糖的抑癌作用。方法用bcl-2单抗进行免疫组化SABC检测,用酶学动力学方法检测酶活性。结果饲喂致癌剂3-甲氨基偶氮苯(3′-MeDAB)的早期(15天),bcl-2蛋白表达已明显增高,实验结束时3′-MeDAB组阳性率为89.5%(17/19),刺五加组为64.7%(11/17)。bcl-2的过量表达与肝组织中γ-GT活性升高和GST活性降低相一致。结论bcl-2蛋白过度表达出现,对实验性肝癌的早期阶段,很可能在肿瘤的发生发展中起着促进作用。
Objective To study observed the expression of bcl 2,γ glutamyl transpeptidase (γ GT) activities and glutathione stransferase (GST) activities during 3′ MeDAB induced hepatocarcinogenesis of rats,and to demonstrate the distinct inhibitory effect of Acanthopanax senticosus to neoplastic proliferation.Methods Immunohistologic staining for MoAb bcl 2 was performed on liver tissve sectious from 3′ MeDAB treated rats.The γ GY and GST activity was determined by enzymatic dynamic method.Results Bcl 2 was expressed in 89.5%(17/19) of the 3′ MeDAB treated rats and in 64.7%(11/17) of the Acanthopanax senticosus treated rats.Bcl 2 over expression was an early event in hepatocarcinogenesis followed by increase of r GT activity and decrease of GST activity.Conclusions The data indicate that the bcl 2 expression is characteristic of the early phase of hepatocarcinogenesis.Acanthopanax senticosus have distinct inhibitery effect on neoplastic proliferation.
