透明质酸对鼻咽癌细胞增殖及裸鼠移植瘤生长特性的影响 被引量:6

摘要 目的研究透明质酸(HA)对鼻咽癌细胞体内外增殖特性的影响。方法以鼻咽癌CNE-2Z细胞为对象,采用增殖实验、裸鼠体内成瘤实验、图像分析及免疫组化等方法,观察HA对癌细胞体内外增殖力的影响。结果增殖实验表明HA在浓度为0.5mg、1mg、2mg范围内,增殖A比值分别为1.89±0.23、2.09±0.26、2.58±0.59,与对照组A值比较有高度显著性差异(P<0.01),且呈量效正相关。HA可使鼻咽癌细胞裸小鼠移植瘤成瘤早,生长快,实验组移植瘤平均潜伏期、倍增时间较对照组显著缩短(P<0.01);裸鼠移植瘤组织切片PCNA免疫组化染色结果显示,实验组细胞PCNA阳性率、阳性面积明显高于对照组,两组比较有显著性差异(P<0.01);平均光密度高于对照组,有显著性差异(P<0.05)。结论HA对鼻咽癌细胞体外增殖及裸鼠移植瘤的生长有明显促进作用。 Objective To study the effect of hyaluronic acid on the proliferation of nasopharyngeal carcinoma (NPC) cell line CNE 2Z and the growth of implanted tumor in nude mice.Methods Cell growth was measured to observe the proliferation of cell culture.CNE 2Z cells were also implanted into the subdermal layer of the nude mice,and HA was injected in the same location,immunohistochemical stain and image analysis techniques was used to study the growth of implanted tumor.Results The proliferation A values of the experimental groups (1.89±0.23,2.09±0.26,2.58±0.59) increased along the increasing of concentration of HA in the range of 0.5 mg,1 mg and 2 mg.There were significant differences between the experimental group and control one ( P <0.01).HA promotes the growth of implanted tumor in nude mice,the latent period and doubling time of transplanted tumor of treatment group was significantly shorted ( P <0.01).PCNA immunohistochemical stain showed that the positive rate and positive area of treatment group were significantly increased ( P <0.01),the mean optic density(MOD) also increased significantly ( P <0.05). Conclusion There was effect of HA in the proliferation of NPC cells and the growth of implanted tumor in nude mice.The results suggests that HA may have an important role in the development of NPC metastasis.
