摘要 采用酸热与超声波结合法,并通过单因素和L(934)正交试验对雨生红球藻(Haematococcus pluvialis)中虾青素的破壁提取工艺进行了优化。结果表明:影响雨生红球藻细胞壁破碎的主要因素顺序为超声波作用时间>盐酸浓度>水浴温度>水浴时间。优化工艺为:超声波(160W~200W)作用2min、盐酸(2mol/L)、水浴(35℃,50min),此工艺条件下用芝麻油提取虾青素,提取率显著提高达到97.4%以上。 ion: In the present study,cell disruption technology of Haematococcus pluvialis using hydrochloric acids combined supersonic wave was optimized by single-fact and L9(3^4)orthogonal test. Results indicated that the most significant factor influencing the cell disruption was the time of ultrasonic wave, followed by the concentration of hydrochloric acid, waterbath temperature, waterbath time. When treating the cell with hydrochloric acids(0.5 mol hydrochloric acid:1 mg dry cells)in waterbath(35℃,50 min), then using(160 W-200 W)supersonic wave treated 2 min,the best extraction yield of astaxanthin was 97.4 % and above.
