摘要
建立环介导等温扩增技术快速检测食源性致病菌不耐热型产肠毒素大肠杆菌的方法。选用不耐热肠毒素(LT)编码基因的6个保守区设计LAMP引物,优化dNTPs浓度、BstDNA聚合酶浓度、反应温度、反应时间等反应条件,评估该方法的特异性和灵敏性。结果显示该方法检测不耐热型产肠毒素大肠杆菌时间短,特异性较好,灵敏性高。
To develop the method of rapid detection for food-borne enterotoxigenic Escherichia coli by loop-mediated isothermal amplification (LAMP)technology. In this study, the primers were designed cording to six conservative regions from the heat-labile enterotoxin(LT)gene. We optimized the reaction conditions of dNTPs concentration, Bst DNA polymerase concentration, reaction temperature and reaction time. The method was valuated for specificity and sensitivity. The results showed that LAMP assay is an extremely and specific method for rapid detection of food-borne enterotoxigenic Escherichia coli.
