高效液相色谱法测定鱼肌肉中的孔雀石绿残留

Determination of residues of malachite green in fish tissues by high-performance liquid chromatography

采用高效液相色谱法测定鱼组织中孔雀石绿的残留量。样品经高速均质后,用乙腈提取,合并提取液,用二氯甲烷再次萃取,有机相浓缩后过酸性氧化铝和PRS复合小柱洗脱,用乙腈定容。以乙腈-乙酸盐缓冲溶液为流动相,高效液相色谱分离,用荧光检测器检测,外标法定量。其检出限为0.75μg/kg,加标回收率为84.0%～90.1%,相对标准偏差RSD为2.98%～6.21%。

A high-performance liquid chromatography（HPLC） method was established for the determination of malachite green in fish tissues.The tissues were blended thoroughly under acidic medium,chloride was used for re-extraction after the former extracts were combined.After evaporation,the organic phase was passed through the acid alumina column and PRS column,then resolved with acetonitrile.Taking acetonitrile-acetate buffer as the mobile phase,the residue levels were determined by HPLC with fluorescence detector.External standard calibration was adopted in the determination.The detection limit was 0.75μg/kg,the values of recovery found by standard addition method were in the range of 84.0%～90.1%,and values of RSD were in the range of 2.98%～6.21%.
