摘要
目的建立骨质疏松模型大鼠骨磷含量测定的钼酸铵紫外分光光度法和骨钙含量测定原子吸收法。方法通过维甲酸诱导大鼠骨质疏松模型。实验结束后处死大鼠,取左侧股骨,高温灰化后,用硝酸对样品消解,再以钼蓝比色法进行骨磷含量测定,原子吸收法测定骨钙含量。结果样品消解完全,样品骨磷含量为139.1~173.9mg/g,回收率在97.8%~100.2%(RSD=1.13%),样品骨钙含量为87.0~228.7mg/g。结论该方法操作简单、快速、准确,可用于对骨质疏松模型大鼠骨磷、骨钙含量的测定。
Objective To establish molybdenum blue colorimetric method to determine the content of bone phosphorus and atomic fluorescence spectrometry to determine the content of bone calcium in osteoporotic rat model. Methods Retinoic acid on induction of osteoporotic model in rats, when the experiment was concluded, the rats were put to death, remove the left femurs, after high temperature ashed the samples, then using nitric acid digested of samples, the content of bone phosphorus was determined by molybdenum blue colorimetric method, the content of bone calcium was determined by atomic fluorescence spectrometry. Results The samples were digested complete, the content of bone phosphorus is 139.1 - 173.9 mg/g, and the sample recovery is 97.8% - 100. 2% ( RSD = 1.13% ). The content of bone calcium is 87.0 - 228.7 mg/g. Conclusions The method was simple, rapid and accurate. Therefore it can be used for determination of the content of bone phosphorus and calcium in osteoporotic rat model.
